So, what would happen?

Referring to this story, what would happen if Rodney Hide was to resign from Parliament, and Winston Peters was to win the subsequent by-election as NZ First leader?

.. and we`d have a new minister for local government. Probably from neither party. Does that mean that Act refuses to support National on Confidence and Supply?

It would not affect any other MP in the House.

And I think National would easily win an Epsom by-election.

Apart from the fact that the PM will have to find a new minister for certain portfolios, which would affect the National/Act coalition relationship.
